Please refer to the product datasheet for known applications of a given elisa kit. We\'ve tested the Mouse Nucleolin (NCL) ELISA Kit with the following immunoassay(s):
Typical Testing Data/Standard Curve (for reference only)
Typical Testing Data/Standard Curve (for reference only) NCL.

Principle of the assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to nucleolin. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to nucleolin. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain nucleolin, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of nucleolin in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Samples: Mouse tissue homogenates, Cell lysates and other biological fluids
Detection Range: 0.156-10ng/mL
Sensitivity: The minimum detectable dose of nucleolin is typically less than 0.059ng/mL. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined by adding two standard deviations to the mean optical density value of twenty zero standard replicates and calculating the corresponding concentration. Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level nucleolin were tested 20 times on one plate, respectively. Intra-Assay: CV<10%
Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level nucleolin were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of nucleolin in mouse tissue homogenates, cell lysates and other biological fluids. Mouse Nucleolin (NCL) ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
