Principle of the Assay: Tetanus Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing Tetanus antigen and an Tetanus Ab-HRP conjugate. The assay sample and buffer are incubated together with Tetanus Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Tetanus Ab concentration since Tetanus Ab from samples and Tetanus Ab-HRP conjugate compete for the Tetanus antigen binding site. Since the number of sites is limited, as more sites are occupied by Tetanus Ab from the sample, fewer sites are left to bind Tetanus Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Tetanus Ab concentration in each sample is interpolated from this standard curve.
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Sensitivity: 1.0 mg/L
Assay Type: Competitive. Intended Uses: This Tetanus Ab ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat Tetanus Ab. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Rat Tetanus antibody ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
