RDH5 encodes an enzyme belonging to the short-chain dehydrogenases/reductases (SDR) family. This retinol dehydrogenase functions to catalyze the final step in the biosynthesis of 11-cis retinaldehyde, which is the universal chromophore of visual pigments.
The sequence of the cDNA encoding p32 revealed that it is a member of the superfamily of short-chain alcohol dehydrogenases. The authors determined that transcripts of the gene were specifically expressed in retinal pigment epithelium. They also determined that p32 acts as a stereospecific 11-cis retinol dehydrogenase in the presence of NAD+ and that it catalyzes the final step in the biosynthesis of 11-cis retinaldehyde, the universal chromophore of visual pigments.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate RDH5 in samples. An antibody specific for RDH5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRDH5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RDH5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RDH5 bound in the initial step. The color development is stopped and the intensity of the color is measured.
