Typical Testing Data/Standard Curve (for reference only)
Intended Uses: The P53 ELISA has been designed for the precise measurement of P53 in human serum, plasma or cell culture supernates. For research use only.
Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. An analyte-specific polyclonal antibody is pre-coated onto 96-well plates. The biotin conjugated second antibody is used as detection antibody. During the first incubation the standards and samples react with the analyte-specific pre-coated antibody. In a second incubation the biotin conjugated detection antibody completes the sandwich. After washing with wash buffer the TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiterplate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample
Background/Introduction: P53, also known as protein 53 or tumor protein 53, named in reference to its apparent molecular mass: It runs as a 53-kilodalton (kDa) protein on SDS-PAGE. Human p53 is 393 amino acids long and has seven domains. It plays a role in conserving stability by preventing genome mutation, as such, p53 has been described as "the guardian of the genome". p53 also has many mechanisms of anticancer function, and plays a role in apoptosis, genomic stability, and inhibition of angiogenesis.
Samples: Human Serum, Plasma Or Cell Culture Supernates
Assay Type: Sandwich
Detection Range: 156 pg/ml-10,000 pg/ml
Sensitivity: <10 pg/ml. PAI-1 / Serpin E ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit. The following patways have been known to be associated with this gene. Adipose Tissue, Blood, Brain, Connective Tissue, Heart, Kidney, Liver, Lung, Muscle, Vascular tissues are correlated with this protein. PAI-1 also interacts with the following gene(s): PLAT, PLAU, PLAUR, PLG, SMAD3, SMAD4, TGFB1, TP53, VTN. Atherosclerosis, Cardiovascular Diseases, Fibrosis, Heart Diseases, Hypertension, Inflammation, Necrosis, Nervous System Diseases, Obesity, Thrombosis are some of the diseases may be linked to PAI-1 / Serpin E ELISA Kit.
