Principle of the Assay: The Human IL-17 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-17 in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates. This assay employs an antibody specific for Human IL-17 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-17 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-17 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-17 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: IL-17 is an inflammatory cytokine produced primarily by an unique lineage of CD4 T cells that plays critical roles in the pathogenesis of multiple autoimmune diseases. Interleukin-17 is expressed by activated T cells and is 57% identical to the 17- to 26-kD secretory glycoprotein encoded by gene 13 of the herpesvirus saimiri (HVS-13). IL17 induces nuclear factor kappa-B and the expression of IL6, intercellular adhesion molecule-1, granulocyte macrophage colony-stimulating factor,and prostaglandin E2, as well as the maturation of CD34 positive hematopoietic precursors into neutrophils. Anti-IL17 antibodies significantly inhibited osteoclast formation induced by culture media of RA synovial tissues.
