The Human IL-10 n/a (Catalog #MBS355300) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The MBS355300 ELISA Kit recognizes Human IL-10. MyBioSource\'s IL-10 can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA).For quantitative detection of IL-10in human serum, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the IL-10 n/a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The IL-10 n/a product has the following accession number(s) (GI #21492460) (NCBI Accession #NP_659579.1). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Typical Testing Data/Standard Curve
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-10polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-IL-10polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-10amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-10can be calculated.
Background/Introduction: Interleukin-10 (IL-10 or IL10) also known as human cytokine synthesis inhibitory factor (CSIF), is an anti-inflammatory cytokine. In humans, IL-10 is encoded by the IL10 gene, which is located on chromosome 1 and comprises 5 exons. The IL-10 protein is a homodimer, each of its subunits is 178-amino-acid long. It is primarily produced by monocytes. IL-10 is a cytokine with pleiotropic effects in immunoregulation and inflammation. It could downregulates the expression of Th1 cytokines, MHC class II antigens, and co-stimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production.
Assay Type: Sandwich
Detection Range: 7.8 pg/ml-500 pg/ml (body fluids, tissue lysates or cell culture supernates) 3.4pg/ml-250pg/ml (human serum, plasma).
Sensitivity: < 0.5 pg/ml. IL-10 Human ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
