Introduction: Immunoglobulin M, or IgM for short, is a basic antibody that is present on B cells. It is the primary antibody against An and B antigens on red blood cells. IgM is by far the physically largest antibody in the human circulatory system. Because IgM is a large molecule, it cannot diffuse well, and is found in the interstitium only in very low quantities. IgM is primarily found in serum; however, because of the J chain, it is also important as a secretory immunoglobulin. Due to its polymeric nature, IgM possesses high avidity, and is particularly effective at complement activation.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with human IgM. Standards or samples are then added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) -conjugated antibody preparation specific for human IgM, mix well and incubated. The more the amount of human IgM in samples, the less HRP- conjugated antibody preparation specific for human IgM bound by pre-coated human IgM. Then a TMB (3,3\',5,5\' tetramethyl-benzidine) substrate solution is added to each well. And the color develops in opposite to the amount of human IgM in the sample. The color development is stopped and the intensity of the color is measured.
