Typical Testing Data/Standard Curve (for reference only)
Intended Uses: This GPI ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine GPI. This ELISA kit for research use only!
Principle of the Assay: GPI ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GPI antibody and an GPI-HRP conjugate. The assay sample and buffer are incubated together with GPI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GPI concentration since GPI from samples and GPI-HRP conjugate compete for the anti-GPI antibody binding site. Since the number of sites is limited, as more sites are occupied by GPI from the sample, fewer sites are left to bind GPI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GPI concentration in each sample is interpolated from this standard curve.
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
Assay Type: Competitive
Detection Range: 1.0-25U/L
Sensitivity: 0.1U/L. Canine Glucose 6 Phosphate Isomerase ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
